          Roundup will kill anything "green" that it touches, it helps if the plants are actively growing, and it needs time (2 weeks say) to translocate to the roots to kill the plants. Annual weeds = easy, pernicious perennial weeds like Bindweed and Ground Elder are harder. If the plot is covered in weeds then aim to treat twice, the second time after a fortnight or so to treat anything you missed on the first pass, and anything still growing. The further we move into Autumn the less effective the "kill" will be, and might be better to leave until Spring when the weeds are actively growing again.

          The other consideration is what is the risk of something like Roundup in proximity to food crops, or on soil / weeds where food crops will then be grown. I put Roundup on my patch at the outset, to kill the rough grass that had been there for donkey's year prior, but I decided not to use any chemicals thereafter.

          Roundup (specifically the latest Monsanto branded formulations) is no long classes as Hazardous; previously, and in the case of all?? generics, the adjuvants in the product (the things that make it stick to the leaf, migrate through the cell wall, and translocate in the plant) were much more toxic that the active ingredient (Glyphosate) itself, but Monsanto have come up with some alternatives which are much less toxic. That's good for wildlife, particularly aquatic, and potentially also for the person applying it "when accidents happen". I'm pretty sure that Roundup Ultra 3000 is not one of the newer formulations, but that said it won't be any more toxic / hazardous than what has been previously used for decades.

          I take the view that Roundup has been used for decades, is used in prodigious quantities in agriculture and horticulture and that any serious consequences would have come to light by now ... but ... the tobacco companies managed to hide the fact that they were killing their customers for decades ... so who knows for sure. I only use it, other than that first-time when creating my veg patch, on paths and the ornamental areas of my garden.
